چکیده

By JAMES G. WHITE AND WILLIAM Knivrr T lIE UTILIZATION of adenosine triphosphate ( ATP) by human platelets during clot formation and retraction is important to the functioning of these cells in clotting physiology.’ 2 Recent developments in histochemical and electron microscopic methods have permitted identification of enzymes hydrolyzing ATP at the fine structural level.t We have applied these combined technics to the localization of platelet adenosine triphosphatase ( ATPase). Enzyme activity was found on platelet surface membranes and in intraplatelet organdIes. In the course of this investigation, ATPase activity was also observed in other blood cells. Enzyme reaction product occurred on red cell and lymphocyte surface membranes, and in lymphocyte mitochondria and nucleoli.

The fine structural localization of adenosine triphosphatase activity on the taste bud in the fungiform papillae of the rat.

The localization of Mg++ activated adenosine triphosphatase (ATPase) activity in rat fungiform taste buds was demonstrated by electron microscopic histochemistry. Reaction product was found on the cell membrane of the taste cells, but not within their cytoplasm. Especially strong reaction occurred in the taste pore. Besides, enzymes were observed in axon-Schwann cell and axon-type I cell inters...

Selective Inhibitory Effect of Adenosine A1 Receptor Agonists on the Proliferation of Human Tumor Cell Lines

Background: In this study, the effects of three structural analogues of adenosine upon proliferation of human tumor cells were investigated. Previous research showed a cytotoxic effect of adenosine via A3 receptor and A1 receptor and sometimes this effect was receptor independent. The researches showed a differential cytotoxic effect of adenosine and its A3 agonists on cancerous cells, while ot...

Induction of apoptosis in human tumor cell lines by platelets

Introduction: It has been reported that platelets can eradicate tumor cells in vitro, although the mechanism of this effect has not been determined. The effect of platelets on the induction of apoptosis in tumor cells is largely unknown. Materials and methods: To investigate this effect, two human hematologic cell lines, K562 and Daudi, were independently faced with unstimulated and thromb...

The Fine Structural Localization of Adenosine Triphosphatase in the Small Intestine, Kidney, and Liver of the Rat

The distribution of the reaction product of a staining method for adenosine triphosphatase (ATPase) in rat small intestine, kidney, and liver was studied with electron microscopy. Several procedures were tried but the best results were obtained from tissue that had been quenched in liquid nitrogen, sectioned at 25 micro in a cryostat, fixed for 30 to 90 minutes at 4 degrees C in formalin-sucros...

Metabolism of adenine nucleotides by ectoenzymes of vascular endothelial and smooth-muscle cells in culture.

1. Pig aortic endothelial and smooth-muscle cells in culture rapidly catabolize exogenous ATP, ADP or AMP. 2. In both cell types catabolism is due to Mg2+-stimulated ectoenzymes. 3. Inhibition and substrate-specificity studies suggest that both cell types possess three distinct ectonucleotidases, namely nucleoside triphosphatase (EC 3.6.1.15), nucleoside diphosphatase (EC 3.6.1.6) and 5'-nucleo...
